Abstract

Approximately 100 proteins representing the most highly populated folds will be prepared and their ROA spectra measured, together with many proteins and viruses from outside sources, to create a large database from which information about the aqueous solution structures of proteins, including the coat proteins of intact viruses, may be deduced using pattern recognition methods. The many structure-sensitive bands in protein ROA spectra will provide information about the fold itself in addition to secondary structure class. Information about conformational elements present in natively unfolded proteins will also be obtained, as well as conformations of aromatic side chains. The application of ROA to membrane proteins will be explored.
