Abstract
The objective is to gain a structural and mechanistic understanding of the ADC synthase reaction converting chorismate into ADC, and the ADC lyase reaction to form p-aminobenzoate, with complementary studies on the conversion of chorismate to ADIC and then on to anthranilate, catalysed by the bifunctional TrpE. The mechanisms will be dissected using mutagenesis, kinetics, inhibitor and structural studies, including attempts to alter the product specificity. The mechanism of inhibition by 2- fluorochorisimate will be determined. The structural and mechanistic findings will be assimilated into the design of novel inhibitors which will be tested for anti-microbial activity. The studies will be extended to enzymes from M. tuberculosis.
