Abstract

We have recently reported the sequence of a gene from Rhodococcus sp. encoding a P450RhF which is of unique primary structural organisation: analysis of the translation product revealed that the P450 is fused to a reductase domain at the C-terminus which displays sequence conservation to dioxygenase reductase proteins. Ethoxycoumarin was found to be a substrate for P450RhF, when over-expressed in E. coli. Fused P450 monooxygenases are of great interest for mechanistic studies, in particular studies of electron transport. In addition, they have great potential in the application of biohydroxylations to organic synthesis. The aim of the present proposal is to purify and characterise this novel enzyme.
