Abstract

Eukaryotic initiation factor 4B (eIF4B) mediates binding of mRNA to the 40S ribosomal subunit during translational initiation and interacts with the polyadenylate binding protein (PABP), thereby increasing the efficiency of re-initiation. We aim (i) to determine the structure of the N-terminal domain of eIF4B which binds PABP and makes sequence-specific interactions with RNA; (ii) to probe the fine details of the RNA binding specificity of eIF4B; (iii) to determine the structure of a complex between the N-terminus of eIF4B and the C-terminal domain of PABP, thus revealing the structural basis of their interaction; (iv) to probe for the presence of structural elements in the C-terminus of eIF4B.
