Abstract

This proposal is to analyse a bacterial dioxygenase namely 2,4- dihydroxyacetophenone dioxygenase (or DAD) which is involved in the decomposition of aromatic compounds from plants. This dioxygenase is of interest since it catalyses the cleavage of an aliphatic side-chain rather than the more usual aromatic ring fission and it sequence shows no similarity to any other protein. Our objectives are to determine its three-dimensional structure by X-ray crystallography and to analyse complexes of the enzyme and site- directed mutants for an improved understanding of its catalytic mechanism. There are implications for the removal of aromatic pollutants in the environment and for the production of chemicals by biotransformations.
