Abstract

We propose to study the conformational changes in the epidermal growth factor receptor (EGFR) during signal transduction that initiates signalling cascades leading to cell growth and division. Receptor conformational changes are masked in observations of ensemble- averaged populations. Hence, we propose to undertake this research by collecting time-resolved single-molecule fluorescence spectroscopy and imaging data on a physiologically relevant group of receptors in the cell. Single-molecule observations not only provide structural information on changes in the distance and orientation of receptor domains, but also allow the determination of the degree of structural heterogeneity within an ensemble to reveal individual stochastic events.
