Abstract

Recently, we were first to apply protein semi-synthesis to the assembly of O-linked glycoproteins. We have also developed methods for the solid-phase synthesis of thioether-linked N-glycopeptide mimics as thioesters suitable for a chemoselective chemical reaction known as native chemical ligation. Herein we propose a new strategy for the assembly of N-linked glycoproteins and glycoprotein mimics. This process involves joining small synthetic peptides containing the sugar structure of interest to the remaining portion of the protein backbone, which is purified from bacteria. Study of these products may contribute to our understanding of the role of protein glycosylation and provide a semi-synthetic route to glycoprotein therapeutics.
