Abstract

We will exploit recent advances in solid state NMR technology, synthetic chemistry and overexpression and mutagenesis of bacterial nucleoside and drug (NupC, NupG) and multidrug or antibiotic (NorA) membrane transport proteins for the following objectives: (1) determine the conformation(s) and mobilities of ligands bound to the proteins; (2) define the ligand-protein interactions and the architectures of the binding sites; (3) define the partition of lipophilic substrates between solution, membrane and protein; (4) illuminate conformational changes in the proteins occurring upon ligand binding. The application of established and leading-edge solid state NMR techniques to understanding ligand binding in particular, and membrane proteins in general, will be advanced.
