Abstract
We aim to determine, by X-ray crystallography, the three- dimensional structure at atomic resolution of at least one membrane transport protein of the Multi Facilitator Superfamily (MFS). We will apply a high throughput crystallisation approach using various MFS proteins. We have already overexpressed and purified 10 MFS transporters, and instituted a protein factory to produce each protein in 2-25 mg batches regularly. To achieve the throughput required, initial crystallisation screening will be automated. In preliminary trials, we have already obtained crystals from 4 different MFS proteins including the one diffracting up to 6 Angstrom.
