Abstract

Combinatorial libraries will be created that will allow rapid determination of the substrate specificity of a proteinase enzyme. Two different types of positional scanning libraries will be created. The first will build on past work with proteinase inhibitors based on peptides derived from the reactive site of Bowman-Birk Inhibitor. These will permit inhibitory sequences to be defined, which in turn will provide information about preferred substrate requirements as these inhibitors are substrate mimics. The second approach will be to generate libraries of peptides that incorporate the fluorogenic amido methyl coumarin group. Iterative deconvolution of these libraries will permit optimum sequences to be defined.
