Abstract

The azinomycins represent a highly unusual class of DNA interstrand cross-linking (ISC) agents which display potent anti-cancer activity. It is known that the epoxide and 1-azabicyclo[3.10]hexane rings are responsible for ISC formation, however, full mechanistic details of the process remain unclear. Under this programme, we will use our expertise in the synthesis of this class of agent to produce novel azinomycin-like compounds designed to address specific mechanistic questions. These compounds will be evaluated in a range of chemical, spectroscopic and biological assays including time-dependent experiments. The details of the mode of action of the azinomycins will be revealed by these investigations.
