Abstract

The enormous impact of the X-ray analysis of the ribosomal subunits from Haloarcula marismortui and Thermus thermophilus is the culmination of 35 years of extensive work. This provides an excellent starting point to address the conformational changes that accompany the various functional states of the ribosome during its cycle. To address these changes we will apply MS techniques developed to capture transient states. Specifically we plan to: (i) Examine the conformational changes that occur in the presence of elongation factors and the nascent chain through controlled dissociation of the intact particle and hydrogen exchange methodologies. (ii) Use MS to enrich samples of the ribosome carrying the nascent chain for subsequent analysis by cryo-EM. (iii) Monitor protein synthesis in real-time at the level of single ribosome assemblies. (iv) Examine the yeast ribosome.
