Abstract

High sensitivity fast atom bombardment, electrospray and laser desorption mass spectrometry screening strategies will be used to compare the glycosylation repertoire of normal and knockout mice, the latter lacking glycosyltransferase and glycosidase enzymes that provide key branching and terminal structures. The proposed work is part of an international collaboration addressing fundamental aspects of protein-carbohydrate structure/function relationships. In complementary work aimed at elucidating the roles of protein-carbohydrate interactions in the immune system we will screen for glycosylation differences in resting and activated T and B cells.
