Abstract

The nucleocytoplasmic transport of mRNA in higher eukaryotes involves a large complex of proteins including Tap, p15, REF, various nucleoporins, E1BAP5, hGLE2, DEK, Y14 and Ran. We have developed expression/purification systems for the core components of this complex. We will initiate crystallisation trials with the core complex and develop expression systems for additional components of the complex. Using the assembled core complex we will identify a high affinity RNA substrate for crystallisation experiments and develop high level expression/purifications systems for this RNA molecule. Cocrystallisation of the core complex with other components will be attempted as they are produced in the project.
