Abstract

The interaction of small molecules with rRNA is timely in view of the recent structural determination of the ribosome. The aim of this research proposal is to synthesise a library of stereoisomeric aminoglycosides (which have both the ring III of the neomycin class and rings III/IV of the kanamycin class) and to investigate the molecular basis of their interaction with RNA using surface plasmon resonance. The stereoisomeric ligands will probe regions of conformational space which are not available to natural aminoglycosides. Ultimately, similar libraries of compounds may be used to select specific RNA sequences which may allow the elucidation of rules which relate ligand structure to RNA binding sequence.
