Abstract
Phosphoinositide (PI)-derived second messengers are generated by both PI kinases and 5-phosphatases. The latter transform the major product of PI 3-kinases into another second messenger thus making these phosphatases distinct players in cellular signalling in their own right. We aim to design and synthesize new inhibitors, agonists, cross-linkers and immobilised ligands by rational modification of the inositol polyphosphate head-group molecular framework. This will not only provide the means to investigate the enzymology of the phosphoinositide 5-phosphatases, but will facilitate the production of custom-made inhibitors for most of the enzymes and potential binding proteins.
