Abstract

The aim of this proposal is to explore the design, synthesis and screening (in vitro and in silico) of combinatorial libraries based on inhibitors of folate-dependent enzymes. Synthetic efforts will be directed towards developing new methods to prepare libraries of potential inhibitors based on a pyrido- or pyrrolo[2,3-d]pyrimidine heterocyclic motif so that leads generated by screening (in silico) may be synthesised and tested (in vitro). The synthesis of targeted libraries of potential folate-dependent enzyme inhibitors represents a new way of analysing and developing these agents, directed towards accelerating the drug discovery process.
