Abstract

Circular dichroism spectroscopy (CD) is an important technique for assessing secondary structures, folding pathways, and molecular interactions of proteins. However, secondary structural analyses based on CD data for membrane proteins have been limited in their effectiveness due to the inappropriateness of existing reference databases which have been derived from soluble proteins. This project will: 1) collect high resolution CD spectroscopic data on the synchrotron radiation (SRCD) station at Daresbury for the membrane proteins whose 3D structures are known from crystallography; 2) create a new reference database of secondary structures and CD spectra and a website with algorithms that can be used for empirical analyses of the structure of other membrane proteins; and 3) use this database to examine unique spectral features found in membrane proteins.
