Abstract

The structural basis for carbohydrate recognition by siglec-7 and related siglecs of the immune system will be investigated. Siglec-7 and sub-domains will be crystallised in the presence and absence of carbohydrate ligands. Using structure-guided site-directed mutagenesis, residues will be identified that are important in the novel specificity of siglec-7. Surface plasmon resonance on Biacore will be performed to obtain real time binding kinetics with a range of oligosaccharide ligands. A specific siglec-7 inhibitor and rationally designed derivatives will be synthesised, together with a novel panel of sialic acid derivatives that will be used in inhibition assays, biological studies and co- crystallisation experiments with siglecs-1 and -7.
