Abstract

The primary objective of this project is to use xylan-binding carbohydrate binding modules (X-CBMs) as model proteins to dissect the molecular basis for protein-carbohydrate recognition. X-CBMs exhibit ligand selectivity, in that they bind specifically to beta1,4-linked xylose polymers within the complex polysaccharide matrix that comprises the plant cell wall, but also exhibit sufficient flexibility to accommodate the residues that decorate the xylan backbone. Resolution of the structural basis for the ligand specificity of these proteins will provide important insights into both the specificity and flexibility of protein-carbohydrate molecular recognition. To achieve these objectives we will solve the three dimensional structure of five X-CBMs in the presence and absence of linear and decorated ligands, and use a rational design approach to investigate the functional importance of key residues that interact with the different ligands.
