Abstract
Sugar-binding C-type lectin-like domains (CTLDs) mediate cell adhesion, protein turnover and neutralisation of pathogens. Sequence comparisons, 3-dimensional structural determination, site-directed mutagenesis and functional arrays identify constellations of amino acid residues that determine selectivity of CTLDs for different saccharide ligands. We propose to undertake an analysis of the human genome to generate and make available an annotated database of CTLD- containing proteins. Annotation and classification of these domains will help to focus attention on CTLDs most likely to bind sugars and will provide guidance in the formulation of hypotheses about the role of sugar recognition in the biological functions of these proteins.
