Abstract

Inhibitor molecules against the gas-gangrene alpha-toxin will be designed, synthesised and tested. X-ray crystallography will be used to help understand the molecular basis of improved inhibition and to guide the design process. Structural studies on site-directed and chimeric constructs will assist in the understanding of how the alpha-toxin interacts with phospholipids. X-ray analysis will also show how the architecture of related phospholipases C is related to differences in their toxicities and substrate specificities. (MOD Joint Grant Scheme)
