Abstract

X-ray crystallography will complement ongoing cryoelectron microscopy studies to reveal the macromolecular structure of photosystem II at high resolution. This membrane complex is composed of more than 25 different protein subunits and catalyses the thermodynamically difficult reaction of water oxidation using light energy. Crystallisation trials will be conducted on an individual proteins, sub-core, core and supercore complexes of PSII isolated from spinach, the green alga Chlamydomonas reinhardtii and the thermophilic cyanobacterium Synechococcus elongatus, for which there are already biochemical expertise and emerging structural models ranging from 30 to 8A or better.
