Abstract

A combination of site-directed mutagenesis, time-resolved and steady-state spectroscopy, physiological studies and X-ray crystallography will be used to explore possible mechanisms for achieving coupled reduction and protonation of ubiquinone in a membrane-bound electron transfer protein. This will be done through the construction of a site for ubiquinone production at the QA position in the reaction centre from Rhodobacter sphaeroides. The project will also explore the application of natural selection as means of activating and optimising new mechanisms of ubiquinone reduction and protonation via the L- branch of cofactors in bacterial reaction centre.
