Abstract

Evidence collected during the past two decades indicates that murine sperm-egg binding is dependent upon the recognition of oligosaccharides that coat the zona pellucida (mZP). Several models have been proposed to mediate this interaction, but the structures of the molecules involved remain enigmatic. Preliminary work from our laboratory has recently provided the first evidence for the presence of the Sda antigen in mZP. We are seeking funding to continue these studies with the objective of rigorously characterising the glycosylation profile of each of the three murine ZP glycoproteins (mZP1, mZP2 and mZP3). This research will be complemented by structural studies of mZP from transgenic mice expressing the human ZP3 gene. This work will lead to a better understanding of carbohydrate-protein recognition in the fertilisation process.
