Abstract

We aim to initiate a detailed structural characterisation of the Reg two- component signal transduction proteins (RegA and RegB), and their interactions with other pathway components, in order to determine how photosynthesis, CO2- and N2-fixation are differentially regulated by the Reg pathway in response to oxygen in Rhodobacter sphaeroides. Purified RegA, RegB and other components will be amplified in E. coli, and subjected to a range of genetic, biophysical and structural methods: (i) to determine the activity, structure and structural changes of these proteins in isolation, upon phosphorylation, and in interactions with their known ligands/proteins; (ii) to obtain the first quantitative data on RegA-RegB interactions; (iii) to identify the Reg-A binding site(s) in the target promoter DNA of photosynthesis genes; and (iv) to identify which of the proposed signals is actually sensed by RegB. The proposed studies, which uniquely involve full length proteins, will also contribute to understanding of two-component function in general (and ultimately to rational drug design).
