Abstract
We have shown that the aphtho- and cardiovirus 2A-mediated polyprotein cleavage is not a proteolytic reaction, but rather a novel manipulation of the translational apparatus. In addition we have shown this mechanism to be a property of other, non-picorna-, virus and cellular sequences. We wish to investigate the mechanism of this reaction using a combination of molecular biological and synthetic organic chemical techniques. This research program will not only give us insights into how 2A interacts with the peptidyltransferase centre of the ribosome but will enable us to use a unique probe into the events and structures involved in peptide bond formation. The proposed program of work leads us into new lines of enquiry, but builds upon both: (a) complementary expertise; (b) already established reagents, techniques and collaborations.
