Abstract

A combination of site directed mutagenesis, biochemical treatments and X-ray crystallography will be used to investigate possible functional roles of a specific interaction that has been identified between an integral membrane protein, the bacterial photoreaction centre, and the diacidic phospholipid cardiolipin. Two conserved basic residues that bind the phosphate head groups of the cardiolipin will be mutated to neutral residues, and the effect on assembly and function of the reaction centre will be examined. Caliometry will be used to examine the effect of cardiolipin binding on stability of the reaction centre. X-ray crystallography will be used to examine the effect of mutations on cardiolipin binding by the reaction centre.
