Abstract

The research will develop further a new and powerful technique to observe and analyse coupled/gated electron-transfer reactions in biological systems, and will examine aspects crucial to the action of both redox-driven proton pumps and Fe-S active sites. Fast scan protein-film voltammetry, supported by stopped-flow kinetics, spectroscopy and molecular biology methods, will be used to study the combined kinetics and energetics of coupled redox reactions of Fe-S clusters, down to time scales greater than 10-3 sec. These include (1) Proton binding at [3Fe-4S]o, and a detailed study of long-range proton transfer to the buried cluster in the crystallographically defined Azotobacter vinelandii ferredoxin and mutant forms, and (2) Unusual reactivities of all-Fe(II) [3Fe-4S]2- clusters, which can undergo very rapid, reversible, 2e/2H+ oxidation likely to involve disulphide-coupling.
