Abstract

Gonadotrophin surge attenuating/inhibiting factor (GnSAF/IF), an ovarian hormone which regulates gonadotrophin secretion, remains to be conclusively characterised. This study will isolate GnSAF/IF from granulosa cell culture- conditioned medium rather than follicular fluid to minimise numbers of contaminating proteins. AIMS: i. To generate N-terminal amino acid sequences and construct synthetic peptides for GnSAF/IF. ii. To generate antibodies to purify GnSAF/IF and screen ovarian granulosa cell cDNA libraries. iii. To confirm the specificity of synthetic peptides and antisera, using GnSAF/IF bioassay and immunoaffinity techniques and subtractive antibody screening. This focused project will provide information for a major research programme, including studies into in vivo effects of GnSAF/IF, production of recombinant GnSAF/IF and development of GnSAF/IF immunoassays.
