Abstract

We will use atomic force microscopy (AFM) to analyse the mechanisms of DNA binding, translocation and cleavage by the prokaryotic restriction-modification enzyme EcoKI, and other enzymes that have a similar mode of action. The application of AFM will allow us to study the dynamics of the enzyme-DNA interactions in real time, an approach that has already provided us with new insights into the way in which EcoKI works. Specifically, we will study the properties of both wild-type EcoKI and a selection of mutants that have modifications in the subunits responsible for sequence recognition, methylation and DNA restriction. In this way we will dissect these distinct functions of the enzyme, and generate an overall view of its mechanism of action.
