Abstract
X-ray structure determination of succinyl-CoA transferase from mammalian (porcine) heart mitochondria. Refinement of the structure to 1.5 angstrom. X-ray structure determination of the catalytic enzyme-CoA intermediate and correlation of the structure with the newly- discovered properties of half site reactivity. Identification of the catalytic groups and the structural basis for substrate recognition by a combination of X-ray analyses, site-directed mutagenesis and comparative studies between the glutaconyl-CoA transferase from Acidaminococcus fermentens and other transferases.
