Abstract
We will study the interaction of C2 domains of protein kinase C and phospholipase A (cPLA2) with lipid bilayers. We will produce recombinant C2 domains, introducing Trp residues at chosen positions for fluorescence studies. We will determine the bound orientation of the domains and determine the energetics and specificity of binding to anionic lipids in the membrane, distinguishing between non-specific (electrostatic) and structurally- specific binding. We will determine residues in the C2 domain important for interaction with the lipid bilayer.
