Abstract

A mechanistic hypothesis is presented for the extradiol and intradiol catechol dioxygenases, in which the number of available iron co-ordination sites dictates the conformation of a bound hydroperoxide intermediate, and hence the choice of reaction pathway. The project will involve: 1) preparation of carba-analogues as mechanistic probes for C-1 vs. C-2 peroxy intermediates in the reaction catalysed by 2,3- dihydroxyphenylpropionate 1,2-dioxygenase (MhpB) from E. coli; 2) monitoring of dioxygenase-catalysed rearrangements of the carba- analogues; 3) use of enantiomerically pure carba- analogues to study the absolute stereochemistry of catechol dioxygenase catalysis; 4) use of synthetic transition metal complexes to test the mechanistic hypothesis.
