Abstract
Flavocytochrome b2 (Fb2) is a lactate:cytochrome c oxidoreductase. Each subunit consists of two domains: one containing flavin and the other haem. The precise details of how the inter-molecular electron transfer from Fb2 to cytochrome c occurs are not well understood. This raises, question 1: Where on the surface of Fb2 is the recognition site for cytochrome c? It is known that electron transfer from flavin to cytochrome c is insignificant, hence, question 2: Why is this electron transfer negligible when the driving force for it is greater than 0.3V? A possible answer is that recognition between these proteins is virtually non-existent. With our success in expressing the flavin domain in E. coli, we are now ideally placed to answer this. In addition we can also address, question 3: Can we design and construct, de novo, an efficient recognition site for cytochrome c on the surface of the flavin domain.
