Abstract

The successful infection of a host cell by a virion involves a series of macromolecular interactions and for the ssDNA Parvoviridae this process is highly dependent on host cell functions. For a number of parvoviruses, including MVM, the capsid protein has been shown to play a crucial role in tissue tropism. Two allotropic variants of MVM, MVMp and MVMi, can serve as ideal models for defining tropism-specific interactions for the Parvoviridae as a result of their pronounced differences in tissue tropism and pathogenicity, despite their 97% identity at the nucleotide level. Our main objectives are to (1) elucidate the nature and extent of protein-protein interactions on the parvoviral capsid surface required for permissive host infection (and pathogenesis) and (2) identify the site on the capsid surface used for receptor recognition, with the MVM variants as models, utilising the method of X-ray crystallography.
