Abstract

The S. guttatum alternative oxidase has been targeted to the Schizosaccharomyces pombe mitochondria where it is expressed in an active form. The aim of this proposal is to probe the structure- function relationship of the plant alternative oxidase by mutating the two conserved cysteine residues proposed to be involved in post- translational regulation and also to investigate the nature of the ubiquinol- binding site. In order to determine which residues are important for activity we will analyse the respiratory characteristics of the altered protein using the yeast expression system.
