Abstract
We aim to elucidate the structural basis of transcriptional activation in the LysR- family of proteins through studies of CysB and CbbR which regulate expression of genes involved in cysteine biosynthesis and the Calvin cycle respectively. Important structural insights into this large family of proteins have emerged with the solution of the crystal structure of the cofactor binding core of CysB. Here we plan to determine structures of this core fragment in complex with inducer and anti-inducers and to prepare crystals and determine structures of (i) the DNA binding domain and (ii) the intact protein in complexes with duplex oligonucleotides containing CysB binding sites. Parallel studies will focus on CbbR which appears to be more amenable to crystallisation in complex with DNA.
