Abstract
Phosphorylation of SpoIIAA by SpoIIAB regulates a key developmental switch in sporulation. We have established the solution structure of SpoIIAA and assigned the spectra. We shall now determine the structure of SpoIIAA-P from a complete set of NMR experimental contraints, paying particular attention to structural changes induced by phosphorylation. We shall also study the interaction of 15N-SpoIIAA with SpoIIAB to identify the SpoIIAA residues most involved in the complex. Furthermore we shall determine the effects in the cell of directed mutations in spoIIAA, chosen from inspection of the SpoIIAA structure, and correlate these with the structures of the mutant proteins to gain an insight into the developmental switch in atomic detail.
