Abstract
Saponins are plant secondary metabolites with potent antifungal properties. A number of fungi can breach this defence by producing extracellular saponin-detoxifying enzymes. We wish to structurally characterise these enzymes and their mode of substrate binding as they constitute attractive targets for the design of specific antifungal agents. Research grant support is sought (a) to complete the X-ray structure determination and refinement of tomatinase, a saponin-detoxifying enzyme from Septoria lycopersici, and to pursue the structure determination of (b) related enzymes from fungi and bacteria and (c) complexes of enzymes and their mutants with substrates and inhibitors.
