Abstract

We plan to address three fundamental questions about the molecular mechanisms of ligand engagement by I-domain-containing integrins by studying the interaction between the mucosal T cell integrin alpha E beta 7 and its ligand E-cadherin. We shall determine: 1. Whether dimerisation of E-cadherin is required and, if so, why? 2. The identity of the site on E-cadherin recognised by the alpha E I-domain. 3. Whether interaction between alpha E beta 7 and E-cadherin exposes a ligand-induced binding site and whether it is possible to block this with a synthetic inhibitor. Our results will be relevant leukocyte integrins in general and provide basic information that is essential for the design of integrin antagonist drugs.
