Abstract
The mismatch-binding properties of the MutS protein from E. coli will be studied by a variety of biochemical techniques. We will examine the structural details of the MutS- DNA binding by preparing truncated derivatives of the protein (less than 30KDa). We will examine the interaction of MutS protein and its truncated derivatives with a variety of mispaired bases, mutagenic lesions and chemically modified bases designed to provide information on the molecular details of mismatch recognition. The overall aim of this project is to gain a thorough understanding of the molecular details of mismatch recognition by repair enzymes.
