Abstract

Oligonucleotides containing 2'-functionalised nucleosides are expected to play an important role in the development of catalytic RNA (by extending the functional group diversity) and in antisense oligonucleotides (by providing resistance to nucleases). The programme will involve i) the synthesis of oligonucleotides containing the 2'-C- functionalised nucleosides. ii) A study of their ability to form stable duplexes, metal ion dependency of their association, their resistance to nucleases and a study of their metal ion-promoted hydrolysis. (iii) A systematic investigation of the ability of the triphosphates of these modified nucleosides to serve as substrates for DNA-dependent RNA polymerases.
