Abstract

X-ray crystallography will be used to investigate the molecular origins of changes in the biophysical properties of bacterial reaction centres that arise following site-directed mutagenesis, or result from chemical treatment of the complex. The project will investigate the changes in protein structure that underlie alterations in the optical properties of the Rhodobacter sphaeroides reaction centre, or that underlie changes in the redox potential of the reaction centre cofactors. The project will also use protein engineering to investigate the mechanism of a new pathway of electron transfer, driven from the excited state of a monomeric bacteriochlorophyll species, that has recently been identified in a mutant reaction centre.
