Abstract

Protein carbohydrate interactions are now regarded as central in biology. Development of therapeutics based on carbohydrates is hampered by a lack of understanding at the molecular level of the processes involved and their contribution to the overall free energy of binding. These processes are solvent exclusion, hydrogen bonding, water rearrangement and van der Waals interactions. Using a combination of micro-titration calorimetery, chemical synthesis, protein crystallography and modelling we will determine the contribution of each of these processes. Successful completion of this project will provide a rational basis for development of new carbohydrate based therapeutics.
