Abstract

Very-short-patch-repair enzymes are small endonucleases with the unique ability to recognise and cleave a pro-mutagenic G:T mismatch base-pair within the context of a specific DNA sequence. In order to understand how these enzymes achieve their simultaneous recognition of a sequence and a lesion, we propose a multidisciplinary project involving detailed biochemical, enzymological and mutagenesis studies of the specificity and mechanism of action of these enzymes, in combination with structural studies of the enzymes and trapped complexes with DNA substrates.
