Abstract

In this application we seek funding for an X-ray generator, operating costs and a postdoctoral assistant to underpin a varied programme of crystallographic-based structural biology projects at Bristol. These include projects addressing the mechanisms by which proteins bind, discriminate and cleave DNA bases (EcoRV, HIV integrase and the papillomavirus transcription factor E2), structured-based drug design (Plasmodium falciparum lactate dehydrogenase and HIV integrase), analyses of protein folding and engineering (antibodies, CD2, LDH and human prion proteins) and probing the basis of allostery in pyruvate kinase. These and other crystallographic projects form part of an integrated approach to biomolecular recognition at Bristol.
