Abstract

Low resolution structures have been obtained of the core particles of bluetongue virus (BTV- 1) and BTV-10. We plan to collect (using synchrotron radiation) a set of X-ray data for BTV-1 to 3.5 Angstroms resolution and for BTV-10 to somewhat lower resolution. Comparison of these two serotypes and with the structure of selenomethionated cores of BTV- 1 will facilitate the alignment of the amino acid sequences with the X-ray structures and lead to refined atomic models. To date, no other structure of such complexity has been solved by X-ray crystallography; we hope to learn how these complex assemblies of molecules are formed and maintained, and to extend the study to core particles of other orbiviruses and to complete virus particles.
