Abstract
We seek to generate improved abzymes and substrates for use in ADAPT. (1) We shall prepare phosphonamidate transition state analogues (TSAs) for cleavage of ortho-nitro and ortho-chlorophenyl carbamate esters and their use as haptens to raise monoclonal antibodies. (2) These hybridoma proteins will be evaluated as catalysts by chemical kinetics using a direct UV assay firstly on simple carbamates and then on self-immolating carbamates. (3) Variation in the cytotoxic entity released by abzyme carbamate cleavage from anilino or phenolic mustards to Actinomycin D will be used to optimise cell kill performance in vitro.
