Abstract
The aim is to determine how the central globular domain of H1 interacts with the nucleosome and how the long, basic C-terminal tail organises internucleosomal linker DNA. (1) Mutagenesis, coupled with a range of functional assays for DNA and chromatin binding, should allow more precise description of the DNA binding sites on the globular domain. (2) H1s with truncation of the C-terminal tail, or with mutated -SPKK- sites, will be tested in a variety of assays, in particular for their ability to interact with AT-rich DNA and to condense chromatin. Major groove contacts of the H1 tail will be identified through labelling with 5- BrU-containing oligonucleotides. (3) The ability of H1 (and mutants, variants etc) to bend DNA will be tested.
